#1 - Infection of sparrows (Passer domesticus) and different mice strains with Lawsonia intracellularis, 33(3):372-378

E-mail: guedesufmg@gmail.com The susceptibility of sparrows (Passer domesticus) and strains of mice (Swiss, BALB/c, C-57 and DB-A) to Lawsonia intracellularis infection was studied. Thirty-two sparrows were inoculated with pure culture of L. intracellularis and eleven received sham inoculum. Feces were collected on -1, 7, 14 and 21 days post infection (dpi) for detection of L. intracellularis by PCR. After 21 days, all sparrows were euthanized and the tissues processed for histology and immunohistochemistry (IHC). One hundred sixty mice of four different strains (n=40, per strain) were used. For each mouse strain, 16 animals received mucosa homogenate from a pig infected with L. intracellularis, 16 received pure culture of L. intracellularis and eight animals received sham inoculum. Two control and four inoculated mice from each group were euthanized on 7, 14, 21 and 28 dpi. Sections of intestine were collected for histologic analysis and IHC and pooled feces were collected for L. intracellularis PCR. None of the sparrows had any histologic lesions characteristic of proliferative enteropathy or antigen labeling by IHC. All sparrow fecal samples were negative by PCR. All mice strains studied had histopathological lesions typical of PE and IHC labeling consistent with L. intracellularis infection, especially those animals inoculated with pure culture. The most severe lesions were observed in DB-A and Swiss mice. Fecal shedding was detected in all mice strains, with peak at 14 dpi. We conclude that sparrows do not seem to be relevant in the epidemiology of L. intracellularis. The results showed variations in the lesions among the four mice strains used.

Trinta e dois pardais foram inoculados com cultura pura de L. intracellularis e onze receberam placebo. As fezes foram coletadas nos dias -1, 7, 14 e 21 após a infecção (dpi) para a detecção de Lawsonia intracellularis por PCR. Após 21 dias, todos os pardais foram eutanasiados e os tecidos processados para a realização da histologia e imuno-histoquímica (IHQ). Cento e sessenta camundongos de quatro linhagens diferentes (n=40, por linhagem) foram utilizados. Para cada linhagem de camundongo, 16 receberam homogeneizado de mucosa preparado a partir de um suíno infectado com L. intracellularis, 16 receberam cultura pura de L. intracellularis e oito animais receberam placebo. Dois camundongos controle e quatro camundongos inoculados de cada grupo foram sacrificados aos 7, 14, 21 e 28 dpi. Seções de intestino foram coletadas para análise histológica e IHQ e amostras de fezes foram coletadas para a realização da PCR para detecção de L. Intracellularis. Nenhum dos pardais apresentou lesões histológicas características da enteropatia proliferativa ou marcação positiva por meio da IHQ. As amostras de fezes dos pardais foram negativas na PCR. Todas as linhagens de camundongos estudadas tinham lesões histopatológicas típicas de enterite proliferativa e IHQ positiva para a infecção por L. intracellularis, especialmente aqueles animais inoculados com a cultura pura. As lesões mais graves foram observadas em camundongos DB-A e Swiss. A eliminação fecal foi detectada em todas as linhagens de camundongos, com pico 14 dpi. Conclui-se que os pardais não são relevantes na disseminação da L. intracellularis. Os resultados mostraram variações nas lesões entre as quatro linhagens de camundongos utilizadas, indicando o potencial risco que os camundongos representam na transmissão de L. Intracellularis.


#2 - Occurrence of pneumonia associated to infection by Mannheimia haemolytica in sheep of Minas Gerais, 29(9):719-724

[Occurrence of pneumonia associated to infection by Mannheimia haemolytica in sheep of Minas Gerais.] Ocorrência de pneumonia associada à infecção por Mannheimia haemolytica em ovinos de Minas Gerais. Pesquisa Veterinária Brasileira 29(9):719-724. E-mail: ecco@vet.ufmg.br This paper describes an outbreak of pneumonia in a sheep herd in the central region of Minas Gerais, Brazil. Clinically, the animals presented apathy, exhibited respiratory difficulty during 2 to 3 days or sudden death. The animals were not medicated and found dead. Grossly, the pulmonary findings were similar in all sheep. The pulmonary cranial lobes and the ventral portion of caudal lobes were consolidated and purulent exsudate streamed out of the airways. In the parenchyma of the cranial lobes there were white slightly prominent multifocal to coalescent areas with 0.2 to 0.5cm in diameter intercalated with dark red areas. Consolidated lesions occupied 70 to 80% of the lungs. Fibrinous pleuritis was observed in sheep 1, 2 and 3. Microscopically, the findings were fibrinopurulent bronchopneumonia with intense hyperemia, areas with intra-alveolar hemorrhage and thickening of interlobular septa with numerous neutrophils, cellular rests and scattering fibrin. Multifocal areas with liquefaction necrosis containing numerous bacterial colonies were observed in sheep 1, 2 and 3. In the cranial lobes of these sheep, there were areas with degenerated neutrophils forming clusters of basophilic cells with alongated nuclei (“oat cells”) associated with bacterial colonies. The histological findings were characteristic of pneumonia caused by Mannheimia (M.) haemolytica. Samples of the cranial lobes were sent for bacterial culture, and M. haemolytica was isolated and identified in all animals. This is the first report correlating pathological findings and the isolation of M. haemolytica as cause of bronchopneumonia in sheep in the country.

E-mail: ecco@vet.ufmg.br O trabalho descreve um surto de pneumonia em ovinos em uma propriedade na região central de Minas Gerais. Clinicamente os animais apresentavam apatia, mostravam dificuldade respiratória durante dois ou três dias ou morriam subitamente. À necropsia as alterações pulmonares eram similares em todos os ovinos. Havia consolidação dos lobos craniais e da parte ventral dos lobos caudais e ao corte fluía exsudato mucopurulento da traquéia e dos brônquios. No parênquima dos lobos craniais havia áreas brancas multifocais a coalescentes com 0,2-0,5cm de diâmetro, levemente proeminentes e intercaladas por áreas vermelho-escuras. Pleurite fibrinosa foi observada nos Ovinos 1, 2 e 3. As lesões de consolidação ocupavam cerca de 70-80% da extensão pulmonar. Microscopicamente, as alterações eram de broncopneumonia fibrinopurulenta com intensa hiperemia, áreas com hemorragia intra-alveolar e espessamento dos septos interlobulares por inúmeros neutrófilos, restos celulares e intensa exsudação de fibrina. Áreas multifocais com necrose de liquefação contendo numerosas colônias bacterianas foram observadas no Ovino 3. Nos lobos craniais dos Ovinos 1, 2 e 3, haviam áreas com neutrófilos degenerados formando aglomerados de células alongadas com formato de “grãos de aveia” associados a colônias bacterianas. As alterações histológicas foram características de pneumonia causada por Mannheimia (M.) haemolytica. Amostras dos lobos craniais de todos os ovinos foram encaminhadas para cultivo bacteriológico e M. haemolytica foi isolada e identificada em todos os animais. Este é o primeiro relato correlacionando os achados patológicos e o isolamento de M. haemolytica como causa de broncopneumonia em ovinos no Brasil.

The total serum (TSP) and plasma (TPP) proteins, gamaglutamiltransferase activity (GGT), serum albumin, alpha, beta and gammaglobulin by electrophoresis in agarose gel and IgG estimated by the zinc sulphate turbidity test were determined. Two-way-ANOVA was used to compare the data in the first age. The age behaviour was analysed through repeated measures ANOVA. Correlations were established between the variables. The passive transfer of immunity was successful in both breeds and the parity of the dam had no effect on the calves’ serum gammaglobulin concentration. The levels of gammaglobulins were higher at the end of the first day of life, and decreased until 60 days. From there on, the increase due to the active production of antibodies was precocious in taurine calves and slower in Zebu calves. At the end of the first day of life, the gammaglobulin was correlated with IgG (r=0,859), TPP (r=0,807), TSP (r=0,811) and GGT (r=0,399). The variation of serum proteins followed the normal pattern throughout the first four months of life, with little differences between taurine and Zebu calves.

The parasitemia with Anaplasma marginale, Babesia bigemina and B. bovis was investigated. Repeated measure ANOVA was used to analyse the age effect on leukocyte counts. Variation of positive neutrophil throughout the age was analysed through Krushkal-Wallis test. Comparisons among breeds were done using the Mann-Whithney test. The white blood cell count increased with age in both breeds, with a decrease in neutrophil and an increase in lymphocyte counts. The neutrophil lymphocyte ratio had been inverted before 15 days of life. The neutrophil oxidative burst was less effective in the newborn calves increasing with age. Limousin calves’ neutrophils had higher capacity to reduce NBT in advanced ages. The observed differences between taurine and Zebu calves had no effects on calves’ health and could be atributed to Anaplasma marginale natural and asynptomatic infection.
